"No" is funny help

I don't know about the rest of you, but we're definitely in that terrible two's stage. He's actually not that bad most of the time, but some of the time... Oy. He's testing boundaries and being defiant and all that good stuff.

Only now, he's developed a habit that is crossing into school and I don't like that at all. His teacher told me today (he's in school two days a week for 4 hours with other 2 year olds) that he screams "NO" at the assistant teacher and thinks it's funny. I totally believe her, because a lot of times when he does something bad at home he'll say it's funny. He was actually sent to the director's office for a chat today because he now has the other kids doing it (and laughing when they do it).

At home I've tried time outs, telling him it's not nice, taking something away if that's what caused it and things like that. He doesn't really tell me no and then think it's funny at home, and apparently he doesn't do it to the teacher, just the assistant. I'm at a loss of how to correct him.

Any ideas? What's working for you guys?
